sup は「下に」、pose は「置く」。話の下に仮に置いてみる、という組み立てから「そうだと考える・仮定する」を表す。I suppose he's busy. は、証拠を示すというより、たぶん忙しいのだろうと考えていること。
Suppose it rains tomorrow. は「明日雨が降るとしよう」で、条件を仮に置いてその先を考える言い方。
I suppose so. は控えめな同意。be supposed to は「〜することになっている」。見込みなのか仮定なのかは、周りの文から読み分けよう。
